Structure activity relationship in sixteen membered macrolide antibiotics.

چکیده
Leucomycin (Kitasamycin) is a macrolide antibiotic having activity against gram-positive bacteria and certain Mycoplasma strains. Comparison of antimicrobial activities of the variuos leucomycin congeners (I-X) indicates a clear structure-activity relationship among them. The acylation of the 3-hydroxy group somewhat reduces the in vitro activity while the acylation of the 4"-hydroxy group of mycarose produces higher activity. The activity is dependent on the length of the acyl

Cladinose analogues of sixteen-membered macrolide antibiotics. VI. Synthesis of metabolically programmed, highly potent analogues of sixteen-membered macrolide antibiotics.
Five novel 3-hydroxyl derivatives of sixteen-membered macrolide possessing 4-O-acyl-alpha-L-cladinose as a neutral sugar moiety were synthesized by using a combination of structurally stable silyl acetal protection and selective hydrogenolysis of a 3"-methylthiomethyl ether to a 3"-OMe group. Several derivatives having n-butyryl, i-butyryl and n-valeryl substituent at the 4"-OH group exhibited ...
متن کاملTAN-1323 C and D, new concanamycin-group antibiotics; detection of the angiostatic activity with a wide range of macrolide antibiotics.
We detected potent angiostatic activity in a MeOH extract from the mycelia of microbial strain S-45628 in the chick chorioallantoic membrane (CAM) assay. The producer was taxonomically characterized as Streptomyces purpurascens. Active principles designated TAN-1323 A-D were isolated and determined to be 18-membered macrolide antibiotics; components C and D are new members of this group, while ...

16-Membered epoxyenone macrolide antibiotics were reductively deepoxidized with dissolving metals such as zinc. Angolamycin and rosamicin which have a methyl substituent at C-12 in the epoxyenone structure were deepoxidized, but not isomerized further to the geometric isomers P2 and P3.
متن کاملVoltammetric study of the partitioning of macrolide antibiotics at the water/nitrobenzene interface. Relationship to the pharmacokinetic profiling of macrolides
The ion transfer of a series of ionised 14and 15-membered macrolide compounds across polarised water/nitrobenzene have been studied by cyclic voltammetry. Fourteen-membered macrolides transfer as singly-charged molecules in the wide pH range, while 15-membered macrolides can transfer as both singly and doubly protonated cations, depending on the pH of the aqueous solution. The results showed th...
متن کاملGastrointestinal motor-stimulating activity of macrolide antibiotics and analysis of their side effects on the canine gut.
For clarification of the nature of the side effects of macrolide antibiotics on the gastrointestinal tract, the motor-stimulating activity of these agents was studied in unanesthetized dogs. The results showed that erythromycin and oleandomycin, the 14-membered macrolides with two side chain sugars combined at C3 and C5 in a glycosidic linkage in parallel, strongly stimulate gastrointestinal mo...
